Suuuper early into our DND campaign (like, the 3rd session or so,) I got OOC flustered at a random dragonborn NPC. After terribly flirting with him IC and rolling well (somehow?) he got himself a date with this dragon and it was a running joke that Finn had a thing for lizards.
Of course, to keep the joke going, Finn flirted with every lizard he could. A lot of those lizards turned out to be very plot relevant. So, in canon, the entire adventure that was our campaign happened because Finn got the hots for some lizards and got himself into their drama. Whoops!
idk if it's just me, but are there any jokes about one of your characters you made canon? like, for example i made a lot of jokes about pearl being an uwu pastel blogger but then actually ended up putting that in her character because, why not
heck, a joke i had ended up fixing a plot hole regarding two characters so sometimes making fun of your characters .... works

My boy shiz didnt have a canon residence yet and im japanese so a lot of my friends pestered me about logan pauls... trip to aokigahara. i joked saying thats where shiz lives and that he hated longan fruit. its made its way into canon now.
what if this random background became a major character in this story. oops, now he is... wouldn't it be hilarious if he became the mc's boyfriend in the near future haha jk... unless...
what if he's a big fantasy nerd and he really believes in a long-gone set of paladin to a point where it's almost like he and the mc are repeating a point in history. HAHA JK UNLESS
I used a random headcanon generator for Yukiya and my OC Chris. With the headcanon being them having met in Club Penguin years ago long before they met each other in real life.
I used it for their plot now, and that's why Yukiya's mascot is now a Penguin and Chris likes to jokingly called him one (Since Yuki can also meant 'snow', so it's kind of fitting too, lol).

with my main ocs being humanizations of tv channels, i have few jokes related to the humanized channels my characters humanize that turned to be canon.
one is with byron and charmy. byron humanizes blaze, charmy humanizes cosmo. one day, my head went "blaze is discount a&e and cosmo is discount lifetime". the two hate each other and when younger, they used to call each other with those nicknames (in a deregatory way).
another is with max. at one time, sony had three different sony max around the world, each other different from the other. the joke became real when i decided that max has an older brother and a younger sister that all go under the nickname of max. maxwell, the older brother, is sony max india, maximilian (the linked max) is sony max south africa and maxine, the younger sister, is sony max hungary.
Britt's entire existence was really just one big pun. Now she and her species canonnically exist in my personal universe with my other characters because I didn't want to waste what turned out to be a cool design.

What if Erin's mother was the leader of the French Mafia? What if she's the reason why Alain's father is connected to them in the first place? Wait,, then what if Alain's father was also Erin's father and he was the result of a one night stand and--
And that's how Erin and Alain became half brothers, a concept I started as a joke but then realized it actually made a whole lot of sense and saved me from created a father for Erin, because now they both have the same father. Neat.
